Remove quotes:

SET Line=%Line:”=%
echo %Line% — will output: C:\Program Files\
Remove a trailing backslash:
SET Line=C:\Windows\
IF “%Line:~-1%”==”\” SET Line=%Line:~0,-1%
echo %Line% — will output: C:\Windows
